摘要(Abstract):
鞭毛蛋白是细菌鞭毛的主要成分,通过聚合鞭毛蛋白单体形成鞭毛的丝状体。鞭毛蛋白的N-末端和C-末端区域相互靠拢形成功能区域,该区域处于鞭毛丝状体的内部;中间区域是序列变异区,暴露于鞭毛丝状体表面。沙门菌鞭毛蛋白具有特殊的抗原性,称为H抗原,是沙门菌分型的依据之一。作为Toll样受体5(Toll-like receptors 5,TLR5)等先天性免疫系统受体的配体,沙门菌鞭毛蛋白是一种强效的蛋白型佐剂,已广泛应用于细菌、病毒和寄生虫的疫苗研制。本文就沙门菌鞭毛蛋白作为载体蛋白在偶联疫苗的应用作一综述。
Flagellin is a main component of bacterial flagella.By polymerization,flagellin monomers form flagellar filament.The N-and C-terminal regions of flagellin are in close proximity and compose its functional domain which is buried in the center of filament.The central fragment of flagellin is a variable region exposed on the surface of filament.Salmonella flagellin is named as H-antigen because of its antigen specificity,which is employed as a basis for Salmonella serotyping.As a ligand of some innate immune system receptors such as Toll-like receptor 5(TLR5),Salmonella flagellin is a potent protein-adjuvant and widely used in the preparation of vaccines against bacteria,viruses and parasites.In this article,the application of Salmonella flagellin as a carrier protein of conjugate vaccine is reviewed.
